Project Summary

In parallel to molecularly bar-coding human EPSCs and single cell genomics for dissecting hematopoiesis, CTSCB will perform similar lineage tracing in hepatocyte and cardiomyocyte development from human EPSCs, using the CRISPR based lineage labelling system and scRNAseq. The knowledge gained from these lineage tracing experiments will be directly tested for optimising the current protocols of producing hepatocytes and cardiomyocytes from human EPSCs in Programme 2. The lineage reconstruction experiments proposed here establish this vital technology for the EPSC system, and thus represent a cornerstone for enhancing the wider uptake and impact of this HKU-developed technology.
